Not sure if this helps but I think Restraunt Bar & Grill have a private room above the dining area!
ah right nice one. I've found this place before for space hire for group things, some really cool venues:

St James Club Manchester is OK if you want to meet customers and get a private room for meetings. Regus have some super offices and rooms by the hour in Manchester (Kings St one is the best) if you need one. St James is ok, for a meeting with a client/customer/whomever it serves its purpose. Its just not a late night drinking den. Having said that they are trying to change their profile and members list. They would probably be very interested if you were bringing in some younger blood (below eighty). BIggest problem I have had with the place is the formal dress code/ Its outdated and archaic.

I see what you're saying. Sounds a tad stuffy!! Portico library looks like it might be a good one for a daytime coffee chat etc.

Soho House are rumoured to be looking for a location in Manchester. Not sure the concept will translate, there has never been a successful members club in Manchester that I'm aware of. People in the city want to see and be seen. The more typical/likely club users live outside of the city centre

Not entirely sure what the OP is looking for, but if it's mainly just somewhere to work / meet, what about the IoD's place in Manchester: http://www.iod.com/your-venues-and-benefits/iod-ve...
