Yeah the gay thing is neither here or there for acting, look at Rock Hudson. But increasing your height by a foot is something else. Heath Ledger as the Joker, that character is now off the movie role list as nobody could ever manage to portray the Joker anywhere near as well as Heath did. I thought that about Nicholson as Joker at one point. Some wag once said AhNold was brilliantly cast in three films, as a knucklehead bodybuilder in Pumping Iron, as a mindless barbarian in Conan, and as an emotionless android in the Terminator.

Difficult to say how well-cast some people are without knowing about the characters beforehand however there have been some people for whom the part they played was so right, you could not imagine anyone else in the role:
Keith Carradine as Hickock in Deadwood.
Jim Kelly as Williams in Enter the Dragon.
Bruce Dern in Silent Running.
Benicio Del Torro in anything he's ever done (totally believable).
Peter Dinklage in GoT.
Brendan Gleeson in Lake Placid and In Bruges.
Eli Wallach in The Good, The Bad, and The Ugly.
Rutger in Bladerunner.
Polly Walker as Atia in Rome.
Swayze in Point Break.
Gary Oldman in anything.

If the original TV series had not set the benchmark for that character then certainly Oldman's performance would have. Good call on Jim Kelly. "Bull-sheet mister Han man. Christopher Walken as Captain Koons in Pulp Fiction. I can't imagine anyone else doing the "Gold Watch" speech.

I was going to suggest Ed Norton/American History X (so close) Christoph Waltz as SS-Standartenführer Hans Landa in Inglourious Basterds Tarantino says it better than I can Tarantino said: Tarantino acknowledged the importance of Waltz to his film by stating: "I think that Landa is one of the best characters I've ever written and ever will write, and Christoph played it to a tee… It's true that if I couldn't have found someone as good as Christoph I might not have made Inglourious Basterds"
