21 and 22 Jump Street are hilarious. A very rare case of a comedy sequel being as good as, or maybe better than, the sequel. Also Life of Brian, Holy Grail, Anchorman, Idiocracy, South Park, Team America, Dodgeball, Dumb and Dumber

+1 for the Big Lebowski. For me, just too many fantastic scenes and superb lines to really isolate any - the whole is a fantastic work that I've re-watched many times and fully intend to re-watch again in the future. There are very, very few films I can say that of.
Agree with the Big Lebowski. Ashes scene? Also, I got a wedding gift of a bathroom rug way back when. The card said: "I thought it would really tie the room together." Amused me greatly. The (ex) missus had no clue. I still have the rug and am still amused by the card many years on. The little things.

Shut the fk up, Donny.

I forgot how funny Despicable Me was, watching it right now and it's still just as good.
Is it the first one where one of the Minions cracks and shakes another to turn him into a glow stick? Loved that bit!!

Ted in particular the scene where Ted beats the guy up and whips him with the radio aerial just reduces me to tears of laughter

Step Brothers, "you can call me Nighthawk".
Agreed. Watched Step Brothers for the first time with my then 14 year old son and we were quite literally crying with laughter.

The drum kit scene especially

Can't believe no one has mentioned BaseKetball yet (trey Parker and Matt stone). The hospital scene in particular...

Not forgetting Bottom Live (not strictly a movie, but full length, so must count). My favourite moment is this:

Watched spy tonight film was funny as hell in every scene with Jason statham in. The begging round the table where he's swearing at everybody had me literally laughing out loud.

Jackass. The scene where they've attached electrodes to each other. I was in the cinema completely unable to breathe through laughing so much.
Some of the Jackass scenes (from all their films) have me uncontrollable.

I find Jackass very 'Marmite' I'm either laughing so hard it hurts or it leaves me totally cold.

Another vote for Naked Gun and the South Park movie (the only film I've ever paid to see twice at the cinema). Also Liar Liar, Jim Carrey absolutely superb and makes me howl every time I see it.

As for TV, some of the scenes in 'Allo 'Allo properly get me every time. Classic comedy.

There's Something About Mary, a film that was on another level for me, the boundaries were moved on. The scene where Ted got his pecker caught in his fly and subsequently rushed through the crowd into the ambulance with Warren shouting behind 'He was masturbating, he was masturbating!'

Airplane, Naked Gun, Police Squad. How can anyone look into the distance and make me laugh out loud is a comedy genius! Take a posthumous bow, Leslie Nielsen. RIP

Haven't seen Austin Powers mentioned yet. Some of the Dr Evil stuff is solid gold.

Dr. Evil: You know, I have one simple request. And that is to have sharks with frickin' laser beams attached to their heads! Now evidently my cycloptic colleague informs me that that cannot be done. Ah, would you remind me what I pay you people for, honestly? Throw me a bone here! What do we have?
Number Two: Sea Bass.
Dr. Evil: [pause] Right.
Number Two: They're mutated sea bass.
Dr. Evil: Are they ill tempered?
Number Two: Absolutely.
Dr. Evil: Oh well, that's a start.
