Am off to NYC on Thurs for a 4 day trip...
This is the thrid time am going and I would agree that you will only spend a limited time in the hotel howver location for me plays a big part...coming back to a central hotel makes it very convieinient to freshen up before you head out for dinner in the evening again.
I found the Westin near Grand Central is good location and reasonably priced for manhattan -
Though I would say unless you are willing to pay through the nose the hotels are not that great in relation to what you pay.
A tip - Try and book a hotel part of the Hyatt group ( they have some excellent locations in Manhattan. They do a price promise where they will match the price of a hotel room PLUS and extra 20% of the lower price. In NYC I easily found a few websites that beat the Hyatt price- I got them to price match and on this trip am staying at the Andaz 5th Avenue for a really good price.
As others have echoed I would personally stay away from Times Square - You have to visit but wouldnt want to stay in that area...
Intercontinental Barclay is a great location but was very disappointed with the hotel...
Must visit places:
Shake Shack
Top of the Rock (better than empire state building)
Grimaldi's
Normas at the Parker Meridian for great brunch
Lukes for Lobster Rolls
Per Se ( though generally for a special occasion as its extortionate pricing)
Shopping - At both Bloomingdales and Macys we managed to get an extra 10% off by being foreign visitors so take your passport with you

I stayed at Hampton Inn, Soho four years ago. The rooms are relatively small but have everything you need and were clean, comfortable, etc. It is a pretty good location, plenty within walking distance and there are cabs and the subway for further a field.Priced Hyatt but bit expensive

Worth a visit is The High Line, an elevated walkway, takes circa an hour or less.

For Steak try Smith & Wollensky, Sparks or Palm/PalmII (ordered by my preference with S&W top).
Plenty of good Italians but some can be "too Italian" such as one I went to where I was sat on a table next to a very well heeled guy dining with a one-eared gorilla who had just "done 5 years" to keep his acquaintance out of reach of the Feds.

Maybe not 7, 8 or more but I could easily do up to 5 nights in New York. Its a brilliant place with loads to see and do. I avoided shopping though, apart from getting dragged through Maceys.
Edit. Not fortunate enough to have a job that lets me do that 20 odd times. Been to many UK places with work but it is in an out, never enough time to look around.

The subway is fine to use in the day but I wouldn't at night. Its the easiest and quickest way to get around - Manhattan might look small but it isn't.
Wish I was going back - love the place. We went to Coney island and walked the Brooklyn Bridge last time we were there - did the twin towers and most of the tourist stuff the first time. Central Park is worth a visit but you'll need a few hours.

The Path runs under the Hudson from WTC to Exchange Place on weekdays, costs $2.25, but you'll need to get a MetroCard (can be used on NYC subway and buses too) as Path doesn't take cash - it's refillable and doesn't cost anything to get an empty card.
20 yards walk from the Exchange Place Path station you get this view..
That's an old 'phone image as it's first one I came across. My Jersey City office is on Exchange Place and we always stay at the Hyatt on the Hudson there too...sometimes difficult to concentrate in meetings when you've got that view out the office window..!
Oh, and in response to comment above, don't be worried about using the subway at night - perfectly safe, safer than London's tube, certainly more law enforcement around...never had a problem in many years.
