In other words, we're not all the same, so test drive for an appropriately long length of time.
I've got an A4 with the sports seats which I rank with my old Rover 800 and Dad's XJ8 for comfort.
I rate it on motorways very highly, the ride could be a little better on really poor surfaces but it's better than expected and it's more to do with the one flaw in a surprisingly refined car considering it's compact dimensions and 4 cylinder engine. I've done 180 miles, a 12 hour day and 180 miles home several times with no aches or pains.
Having changed jobs I tend to do 10 hours commuting a week in it. 2 litre FSI means about 35 mpg average which is acceptable considering no DPF or turbo to worry about and it's now upto 183,000 miles.
The tyre wear is pretty high though about 15K per conti at over £100 each with completely even wear.

Something a little out of the ordinary, but having driven one from New York to Pittsboro, NC (nearly 500 miles in 1 go)recently, I thought it was pretty good.

Whilst I expect the answer is probably something like an S80 or S60, if you wanted to take a punt on something a little bit more leftfield, may I suggest a Jeep Grand Cherokee Overland CRD (2005-2010). I have just returned from the Alps in my 2007 model (with just shy of 125,000 miles on the clock) and it has not put a foot wrong in the year and a bit since I bought it. In terms of motorway driving advantages:
1. It eats miles quietly (once up to speed;
2. is quick enough (215bhp Mercedes 3.0 V6 CRD - so 0-60 in about 9 seconds and comfortable cruise at 70+);
3. It comes as standard with climate control, heated seats, sat-nav, cruise control, a good stereo, automatic wipers, auto dimming mirrors;
4. The seats are comfy; and
5. High viewing position.
It's not the most economical, but it isn't bad on a long drive. On the round trip to the Alps, including going up and down the mountain, I averaged over 29mpg.
You'd get a decent one for £7,000 I'm sure.

Had one as a hire car once and honestly, there were no redeeming features. It's ugly, slow (CDTi), poor rear visibility, seats aren't especially good, it's noisy at speed, the dash is awful, stereo was rubbish and the seats were average at best. Bloody awful thing.

Facelift has DRL's and 'pinched' rear LED lights.
Le Mans spec has custom alloys, 30mm ride drop, MMI-High, interior LED's, cruise..
Can't beat all-weather quattro for motorway snoozing..
