A6, A75, A76 to Glasgow and back

A6 is not worth the effort south of Lancaster. Come off the M6 j36 then Kendal, A6, By-pass Carlisle using M6 j42-44 then A74. After that I'd do B7076 past Lockerbie as it runs parallel to the M74 (option if you get bored or running late) then use the M74 for the run in to Glasgow from j8. You can always try the M77,A77,A74 on the run back but it is further.

I think you meant A76 not A74. The latter wouldn't form any part of that route. It runs east across the Clyde via Bridgeton to Celtic Park and the Chris Hoy Velodrome then on to Mount Vernon and the M74.I wouldn't choose A75/A76/A77/M77 if going to, or from, Glasgow. It's not only further but a LOT slower. The A76 between Dumfries and New Cumnock can be nice if traffic is light but from there to Kilmarnock is nothing special. Also there is less than 20 miles of motorway plus the A75 is the only viable route to the ferry for Northern Ireland from Cairnryan. So you're likely to be dicing with plenty of HGVs between Gretna and Dumfries.
I wouldn't bother. The M74 is pretty open & empty, the B7076 that runs parallel is the old A74 and as you might expect, the surface is a bit cruddy since it was downgraded to a B road.
If you want a blast & don't mind putting off a bit of time, leave the M74 at Moffat then go A701 - A721 - A70 - A721 then either the A73 to the A8/M8 Jct 6 or the back road from Carluke to Garrion Bridge (marked as Mauldslie Road on Google Maps) then A72 to M74 J7. If you're REALLY not in a rush, go A701 - A72 towards Biggar, then A73 towards Lanark then back onto the A72 to M74 J7. The A72 north of Lanark is now all 40mph sadly & goes through garden centre land, so expect big queues of Honda Jazzes on a Sunday.
