Australia road trip - must see/do things?

We're off to Aus for November, with the plan being to fly to Sydney, have a week there visiting friends then pick up a camper van and drive from there to Melbourne, then up into the wine region above Melbourne then over to Adelaide, from where we fly home.

Are there any must-see/do things along the way that people would recommend? Particularly on the Sydney -> Melbourne leg.
