Have you looked in the Chilterns?

Not quite 150m vertical on this one but there's car parks at the top from memory - https://www.strava.com/segments/618739

I must be the most unobservant cyclist living! Never noticed the brewery sign in the 20-odd ascents/descents I've made of Staple Lane.

Very much a wild card idea would be multiple loops of the Leith Hill Octopus, the 8 separate climbs that all cross over at the (road) summit car park. Each full loop is almost 6,000 ft vertical but over 55 miles total due to the flattish sections linking the 'tentacles'. It's been used for unofficial charity events before, but I suspect none have tried more than one loop. It wouldn't be much of a social, or spectator-friendly, event as riders would soon become strung out around the route, which would minimise traffic issues. I expect it's a non-starter - but there's probably a record to be claimed for the most loops between dawn and dusk! https://www.cyclingweekly.com/sportives/sportive-n...
