My L322 Range Rover scores very highly on the comfort front. Six hours driving with no stretching or walking like a cowboy when getting out of the car.
I had a '93 Rover 827 back in the day, and I remember that being very comfortable, too.
The Rover wasn't an anomaly, either. I've owned lots of barges over the years (Jags, BMWs, Mercs, Audis, Fords, Vauxhalls, etc.)
But those are the two that stand out for comfort.

My 03 Merc E320 is a great way to cover ground, spacious, quiet and comfy. My other half has a CLS350 and that's probably a little more refined but again, a lovely way to cover distances.
I may have been in something comfier in my time but those two stand out as the most comfortable in recent times.

Citroen XM
Jag XJ40
Rolls-Royce Silver Spirit
I must admit, the XM was astonishingly comfortable
Conversely, two examples of cars I expected to be comfortable but weren't:-
Audi Q7
Range Rover Sport (both original and current versions)
Admittedly, the NVH / bump-thump suppression was good but the primary ride quality - for a premium product - was shocking.
