Ordinarily, clearing the cache should effectively be the same as CTRL+F5 (for the specific site only) as it forces a full refresh of the page and associated code.

You haven't accidentally switched to mobile view whilst using the developer console (F12) have you?
Open the developer console using F12 and then press CTRL+SHIFT+M.
EDIT: If not then I'm going to assume that it's an extension you've got installed blocking something (like one of your adblocker extensions blocking the desktop CSS) or adjusting the stylesheet on the fly (something like Stylish).
