Rear spindle bearing has worked loose

After yesterday evenings ride down some really bumpy bridleways (sh!t dodging, and fly swallowing
), I noticed that my rear spindle hs started to come loose with some visible side to side movement, and that horrible Raleigh Mustang type noise over bumps.
When I take the wheel out to clean and tighten this later this evening, how tight should I do it up? Is it as simple as undoing the lock nut, tightening and re-locking? Are there eny left hand threads to deal with? Should I grease the bearing at the same time?
Hub is a boggo Deore affair.

Hi mat, I'm assuming that this is a hollow axle set-up with a quick release skewer and doesn't have cartridge bearings. When you adjust one of these hubs you need to leave a trace of play in it, as when you clamp it in the frame the Q.R. skewer will take up the slack.
The only way to do it is through trial and error and the axle is right hand thread. Make an adjustment, leaving a trace of play then test fit it in the frame clamping the Q.R. skewer securely and check for play. Keep making small tightening adjustments until the play just disappears and your done. make sure you fully tighten the jam nut on the axle and the skewer fully at each adjustment.
In order to do a proper job of greasing the rear axle bearings you will need to have the special tools required to take off the cog set, an axle vise, etc.
