What systems are in place to defend against such a missile?

Tricky, at mach 10 you'd only have a few seconds (c.10 from an envelope scibble assuming it was low altitude) of an anti-ship missile being visible before impact, so it doesn't give you much time to plot a trajectory and intercept.

Well if you can't stop the missile you have to find and destroy its launch sites.
