Plasmas need to be 'burnt' in, ie. it sort of matures to optimum condition after 100-200 hours of use.
After that, I'd get yourself onto AV Forums and have a look for your tv and see what other users are recommending as the settings for your screen. What I did with our Panny G10 a couple of years back.

As mentioned, worry about it once you've put a few hundred hours on the set, but in the meantime keep it on Movie/Cinema mode, and knock the brightness & contrast back a few clicks.After that, get a setup DVD, and there's a 1920x1080 (Nokia?) Test Card JPG floating about online.
Between those, you should be able to tweak the settings so you get the optimum levels for your viewing conditions (unlit room at night for example).
