Yep, but better off investing it in different companies to reduce risk, or maybe a tracker fund.Can the £20k annual stocks and shares ISA allowance be invested in one share?

Yes, but presumably you eventually want to have a portfolio of 25 to 30 businesses to achieve a sensible diversification.
If you want to build the numbers up within the annual ISA allowances more rapidly, how about 4 at £5,000 or 2 at £10,000.
Obviously more risk initially, but that depends on the particular businesses.
You might perhaps be talking about a maturing employer share save option scheme. If so, I expect you know that they can be transferred directly to an ISA.
