I have had this sore throat and the feel of swolleness and general discomfort now for just about a week, it's starting to get tiredsome. I don't generally bother the doctor with minor things but I thought 4 days was bit too long to keep these symptoms so went to see him. He said it looks a bit red down there and prescribed me some penicillin (with other stuff in it) to be taken 4 times a day until the course has finished. The thing is, I have been taking the penicillin for two whole days now and it feels no different, how long does it generally take to work?

Antibiotics generally take about 48 hours to kick in, although proven to reduce symptoms of tonsillitis by only 12 hours over no antibiotics. I suspect you have a virus which the antibiotics will have no affect on.

Fluids, rest and simple analgesia will help

3 days to make a difference ime, assuming this is actually bacterial tonsilitis.

If it is viral, the antibiotics won't do much to help.

Gargle with dispersible aspirin - will help with the pain.

Well I woke this morning to find there was much relief (I always found it felt not too bad in the mornings) but I didn't bank on it as it usually gets worse later in the day. I am pleased to say it still is a hell of a lot better, hopefully they are doing the trick.
