It will be used regularly on and off road so just want to make sure that I will be happy with it long term.

Have you driven a manual and a auto vehicle before? If so, which do you prefer?

Auto will not be quite as quick or economical. But it is more relaxing and has certain advantages off road.
Manual will be more fun, more performance and better attainable mpg. And offer more control in certain situations. But with 6 gears will mean you are constantly changing gear.

A lot of off road is down to the driver, so I am willing to sacrifice a small amount of off road ability for the on road advantages.
The auto is really good on a D3, just make sure it's had a fluid change at 80-90k.
Manuals can suffer from clutch failure, quite expensive to fix.
Try both and see how you get on.

Some people can get way over 35mpg out of their auto's so dont write them off as low 20's.

Having driven plenty Disco 3s with both types of gearbox, I much much much much prefer the auto. It's more pleasant, easier, and really suits the whole feel of the car. And as previously said, I'd be surprised if it reduced your mpg by more than 2 or 3 assuming you drive it normally.
The transmission calibration is very good, without ridiculous amounts of torque converter slip; if you've not driven many decent auto gearbox cars, I'd suggest you try one before committing to the manual!

I sort of agree with this. I've been in a few and some friends with auto TDV6's seem to average around 27mpg on the trip.Having driven plenty Disco 3s with both types of gearbox, I much much much much prefer the auto. I personally prefer the manual as it's more fun. But I did find it pain always having to change gear. It would have been so much better with a 5 speed not 6 IMO.
