Calling Jira experts
Discussion
The small start up business i work for was using Trello cards which worked really well, but our investors are keen for us to use more formal tracking tools. They suggested Jira and we have started using this for each of our projects. As a Programme manager, i am keen to see a holistic quick overview of my portfolio of projects.
Can anyone advise me on how best to set this up? I want to see project timelines and milestones at a quick glance across my portfolio.
Also any tips and tricks to help with monitoring projects?

You've mentioned Programme, timelines and milestones.....JIRA isn't for waterfall delivery. Can anyone advise me on how best to set this up? I want to see project timelines and milestones at a quick glance across my portfolio.

As mentioned, JIra is more suited to agile, but the sort of functionality you’re describing is handled by Advanced Roadmaps so look into that: https://www.atlassian.com/software/jira/guides/roa...
Interesting that you opened up describing a small start-up business. Jira, whilst undoubtedly the industry standard rolls-royce, can be a significant overhead, which is fine if you're a large corporate/public sector where you can absorb this overhead, may not be so fine for an SME with limited resource and budgets etc. From my own experience (and this may just be me), trying to do anything above and beyond basic epic/story/ticket management, Jira can slow things down significantly, basic reporting, dashboarding, summaries etc can become time consuming things, at very least to set up initially.
Appreciate you may have no say if this is being dictated from above, but worth taking time out to properly challenge/assess it's suitability before jumping in - just my opinion fwiw.

I've been using Jira + Confluence + MS Project to run change programmes for a good while now. I hadn't heard of Trello but it looks to be fairly well adopted (if their marketing blerb is correct - 1,000,000 users, big firms inc Google etc..)
For your investors to dictate / suggest a PM tool suggests there is something they are not getting today from the firm / project. I think if you could get to the nub of that you would be in a better place because "just using Jira" may not solve whatever their actual issue is.
Is it lack of delivery? Reporting? Issue management? They don't undestand how to use Trello but do know how to use Jira?

Trello is owned by Atlassian just like Jira.
I run Atlassian’s largest solution partner and we’re also partners in a number of other ecosystems like Aha! , Slack, Monday.com and Plandek. I would offer the opinion that many SMBs and startups are looking at Monday.com as something a step up from Trello but not as “heavy” as Jira. Might be worth some investigation for you.

Was very surprised to see Jira mentioned on PH
We've been trying to migrate an old on-prem Jira Software implementation to the cloud and have uncovered a shadow IT community of well over 500 Atlassian accounts using the full gamut of products bitbucket, trello, software, service, work management, confluence etc. all over Europe and trying to onboard them to a proper Atlassian cloud implementation with SSO/MFA is painful.
Atlassian won't get involved in any kind of vendor risk processes, or help you with clarifying their onboarding processes when you're coming from a relatively complex starting position. It is good software though tbf, the organisation are just painful to interact with (or at least that's been my experience).
