If I was to get my MX5 properly alligned and setup, would changing the wheels to a larger 15" (From 14", wider tyre too) require another allignment or would it cause any issues.

Get it done now, or wait for new wheels and then do it?

If the car isn't pulling to one side, and the tyre wear is normal, there's no reason to do anything - the wheels are adequately aligned.

If you have issues though, and are changing size, I would be tempted to do it once the wheels are on. This is only because aftermarket tyres tend to be more variable in terms of conicity and if you do suffer conicity drift the wheel aligner will be more inclined to make an adjustment if they have supplied the tyres.

The above is all correct but if you're in any doubt a full tracking alignment is inexpensive and worthwhile...
