Any V12 engine is expensive to tune, because it has a lot of parts in it! Just the usual "port and cams" is a big expensive job! Best bang for buck is going to be a low blow turbo / centrifugal supercharger job with something like MegaSquirt managing the whole lot.

3 chips would help, 1 for each engine side, and one for the throttle EML ecu.

I'd leave the internals of the engine untouched myself, you're gonna have enough things to deal with in the install / commissioning itself without worrying about engine internals. A decently designed exhaust (headers + low back pressure silencing) and intake (low loss airbox / filtration / dirty side cold air pipework etc) in conjunction with a decent management and calibration should see you healthy gains over the std BMW numbers without much work / worry.

I know you're probably set on a V12, but I can think of many cheaper ways to 450hp.

A crate LS3 will be 500hp as standard for not a great deal more than the supercharger kit alone.
