I'm possibly going to be doing a commute that will take me the following route, getting to the John Charles Centre at about 5pm, so I'm worried about rush hour traffic. I guess I will be going against it, which will be in my favour but how long do you think it will take? I'm only there for 1 hour 15 mins of work, so don't want it to taker much longer han 45 mins (sounds a bit crazy that, but it's good work).

I'd personally go against the traffic all the way into Leeds, then get onto Dewsbury Rd either Armley Giratory then through Holbeck, or past Evening post, turn right and round the back. I end up at John Charles quite a lot and this year the ring road has been awful. Owlcoates and the lights near JCT Porsche really clog it up

Agree with this ^.This is the route you want to take.
I know that side of Leeds well as I do it most nights of the week and that will be your quickest route in terms of traffic avoidance. Make sure to come off at Dunelm Mill after the gyratory and go through Holbeck as Ingram Rd through the tunnel has a habit of fooling you it's clear until you get to the first right hand bend and then you'll find yourself at a crawl for the next 5-10 mins.
As Roy says, Rodley is best avoided as it will be queueing all the way down the hill from Broadway rbt and then you'll have traffic at Dawson's Corner (Owlcoates), Tong Rd lights, Lower Wortley lights and the old Route2 lights at Geldard Rd.
Going down through Kirkstall should flow reasonably well going in that direction and you'll only have a brief stop at Kirkstall Lane, the viaduct lights and the ones under the inner ring rd.
The only bit you might want to experiment with is going up through Beeston. Tempest Road has speed humps the size of small countries so I wouldn't recommend going down there if your motor is lowered otherwise it might not end well for you!

Time wise for the route I've given you, expect 25 mins on a good day, 40+ mins on a bad day.
