Yes, it stands for sports, as in convertible, from the 3000S as does most of the styling and idea of the car. It was a cheap car and cheap fix to save the company - and it worked very well.

Perhaps to differentiate from the 280i (ie Tasmin) it was called 280S, then 290S then S2 and S3. I think the orginal ads said "TVR Convertible" - as did the 3000S!
TVR had used S2, S3 and S4 before - for the Vixen series.
Was there ever a 290i?

It was called the S, then the S2, etc. Quite a good guide can be found here www.pistonheads.com/tvr/s/Default.asp?storyId=3273 The 280S type of name was a legal thing for the log book only.

No, he RV8 based Tasmin Series was well established by the time that the 290i would have been available and the V6s could not find any buyers then.
