I on a Marcos with the standard 3.9 litre V8.

Can I put in the 5 litre using the same engineloom and ECU? Very likely the chip has to be replaced.....

If its a 14CUX then yes it is just a chip change i believe. I don't know the engine setup in a Marcos but you might also need to upgrade the induction side (intake manifold and AFM) to make the best of the engine

But if I buy a 4.5 or 5 litre from a Chimaera/Griffith complete with AFM and intake, this would be ok or not? I have the 14CUX.

I did the 4>5 litre upgrade in a TVR two years ago.
You can buy a 4.5 or 5 litre engine from a TVR, or
V8 Developments can build you a 4.5 or 5 litre engine that is cross bolted and internally balanced, and with a cam designed to give you the performance you want (more torque or more BHP).

I would start with the standard induction, as the need for more is borderline - it depends on what valves you have.
I went the V8D route and did upgrade the injectors, AFM, plenum etc, but am not convinced I needed to, as my heads are stage 3, so the 4.6 valves, not the larger 5 litre TVR valves.
The standard TVR 5 litre has standard induction anyway.

Once the engine is in the car, the only thing you need to get done for it to run is a remap of the chip in the 14 CUX - go to either Mark Adams (Shrewsbury) at www.tornadosystems.com or Joolz (Chesterfield) at www.kitsandclassics.co.uk to have the car mapped on the rolling road.
Or Mark can supply you with a best guess chip.
The only other thought I have is that if you get a totally standard 4.5 or 5 litre TVR engine with totally standard induction, then all you need is the chip from the donor car's ECU.
Or Mark can supply a standard chip.
Or you can get the entire ECU from anyone who has recently upgraded to an aftermarket ECU. The 14CUX is the same in all Chimaeras, its only the chip that varies with engine size.
