Currently got 275/45 R21 Scorpions on my L405, but it's pretty crashy over potholes and ruts, etc. Don't really want to go down to 20" wheels, so was thinking of fitting 275/50 tyres instead, for a bit more sidewall.

Anyone tried this?

You wont have any problem doing this as someone on Full fat has done the same.

My 405 was on 22`s when I got it. It was awful.
I have gone down to 21`s now and the ride is much better. It seems to handle better through the bends too.

I'm running 275 / 45 /21 on mine and there looks to be plenty of room for 50's gaining about an inch in diameter and 3mph over reading on the speedo

A tiny bit less but this means the true speed will actually be bang on, allowing for typical speedo over-read at, say, 70 mph. Just have to remember the margin for error with cameras is now quite small.

Is the same true on L494? Wanting to fit general grabbers and a bigger sidewall would help

I think you can go up to 32" total diameter without worry on the L322 and L405, I think the L494 is the same. My first two Rangies were on lift kits and 32.5 ish tyres. Lovely ride but ever so slightly squishy on the turn. Radials are definitely quieter, with M+S or AT tyres with blocky treads you can get drone between 40-60. I've gone for 275/50/21 All Seasons this time as a compromise.
