Why do you want to run it from the DOS prompt - are you wanting to defrag the Windows system files? If so, use PageDefrag from SysInternals - unzip it, run the program, set it to defrag on boot and reboot the machine. This is a fantastic little utility that I've been using for years to optimise by computers and it really works and it's free!
