In answer to some of your questions...
Yes, you should have it done
They are to do with the valves afaik, I'm sure someone more technical will come along and correct me. Google is your friend for that sort of stuff
Depends where you take it - Gatwick charge £352 for tappet adjustment on top of £936 for a 12/24k service but they are pretty much the most expensive in the country
Some indepedent dealers will do a 12k service and tappets for £500, at least according to the adverts on here

Sorry to take a little off topic, but surely tappets denotes the presence of rockers?
Isn't it the clearance between the rocker arm end and valve top plate that you are setting?
Never understood why TVR don't just use hydraulic lifters and cam lobes pressing directly onto the valves via them... do they actually use a crank> geartrain > cam > pushrods > rockers? (can't find any pics or diagrams) Any reason for this? I know it's good for engine protection because pushrods fail before pistons slap valves etc, but was that really a concern?
Still alot of money though, how hard is it to set the clearances really? Is it just getting to them while the engine is warm more than anything? (engine long way back in engine bay) Remember setting these quite easily on an old 2.9 V6 Ford engine using the same setup timing method...
