If you're after pick up and play with very intense action then Dawn of War is fantastic.
For a more build up slowly over days type game then Medieval Total War 2 is utterly superb, it will take over your life for several months though!
Supreme Commander is apparently the new kid on the block, I've only played it once and it didn't really do anything for me.

Id suggest getting wc3/tft and then playing some dota www.dota-allstars.comit can be massively fun but is hard for beginners to get into because 90% of players are complete pricks and think they are gods gift to the internet.
I'd vote for Supreme Commander, best strategy game i have played in years
follows on from where total annihilation left off (game-wise not plot-wise)
and the ability to zoom right in and out of the map its so useful that it will drive you nuts whenever you play another strategy game if future!

Another vote for Dawn of War, they are bringing out yet another new expansion pack called 'Soulstorm' which will bring the total number of playable 'races' up to 9, and with a very big 'Risk' style campain map.. Oh and it'll also work on most vaguely modern pc's unlike Supreme Commander, which is utterly useless on anything less than a 1k+ pc.
