The OP's kids are too young for boosters, and you'll struggle to get 3 seats across the back of a lot of cars anyway.If hotel - connecting rooms.
Otherwise, I would recommend the caravan (Eurocamp etc) or villa option; somewhere not too hot.
If wife is knackered, doesn't fancy self-catering and funds allow - Mark warner or Sunsail type holiday places (and some Pierre et Vacances) offer creches/kids clubs and babysitting so you can get some time by the pool whilst the little ones are entertained.

I'll hopefully be taking the family (with 2 young ones) away in Sept/Oct. Cost not really an issue (as we've not had an abroad holiday for around 5 years due to building a business, not because we are condsiderably richer than yooouuu!!). Quite fancy a villa near a beach, ideally with use of hotel facilities nearby?
For obvious reasons, flight time to be minimised - and the place to be child friendly (although I'm not sure we'd feel comfortable using an unknown 'nanny').
I'm thinking Spain, South of France, Portugal (Algarve) etc
(Greece, Cyprus too far? too hot? for little ones)
air tickets around 700 -800 with Thomas cooke , Hot but not over the top for the kids,
Plenty of things to do good restaurants ,and far enough away from the touristy Hurghada ( 20km) to avoid the sellers or with in easy reach if you fancy a camel etc.
This year We've travelled , England , France , Russia , Siberia with the little one shes 8 months and planning Egypt in Oct, no problem with the flights or trains ,
We have 3 kids and they have all done long haul. We choose night flights and generally they go to sleep.
Been to France a couple of times in the car but we have found it much easier to get on a plane with them.
It is harder to find big enough rooms but we find hotels with inter-connecting rooms and use it as one big hotel room.

We have 3 boys 8, 6 and 14months we head to France Key camp or similar we have done this for the last 6 years. It's easy it's not my ideal holiday but if they are happy you can relax. You can take there bikes, cricket bats and so on.
We went to Dubai before number 3 came along stayed at the Jumeriah beach lovely hotel but if you ask the boys would you like to go to France camping or the posh hotel it's camping every time.
The best thing before they are of school age we could go for 3 weeks in June for £800 +petrol and a stop over.
I was thinking about taking them to London trying to find a reasonable hotel in central London for 2 adults and 3 kids is expensive.

Have a look at Mark Warner. I can recommend their Lakitira resort for those with kids. They have various options for families with 3 kids including interconnecting rooms (stick the older two in there and the baby with you) or a suite.
And it has the added bonus that you can put some or all in the kids club for a bit to give you and your wife a bit of time with out them if you want
