What clues to it being a replica ?

I did wonder if it was one of those BMW Z3 based replicas but I don't think so.

It looks the genuine article to me.

There can't be that many RHD SWBs around, I wonder if it is this one?

I think the early versions had the roof line / door frame style of the spotted car, so my guess is it's a genuine car. Worth many millions.

There are two BMW based replicas ( see kit car forum) but the windscreen looks too good for those. AFAIK, genuine 250SWBs have either wind-up windows with quarterlights or sliding perspex windows.
There have been a number of re-bodied Ferrari 250GTE etc 'replicas' with windows like the car in question and I think it could be one of those.

I think we can safely assume that as DKE is/was selling it, that it is a Ferrari but as to whether it is a rebodied one as you describe, dunno.

I dont know either. A number of the Ferrari specialists have sold Ferrari 250 based replicas of the GTO (and SWB?); they were clearly advertised as what they were - no intention to hoodwink anyone. DK Engineering have a very nice Daytona Cobra recreation for sale at the moment for example.

cartell and DVLA both say Ferrari.

First registered September 1960, 2,953cc. MOT history (does it really need one, legally speaking?) shows what appears to be an odometer change in 2009/2010. Old odo was reading 100,273 at it's last MOT, with 3,157 on it's new one. It's had a few advisories too.

A little digging suggests it's the V12 Colombo engine, from the 250 GT LWB/GTE or 250 GT SWB/GTO/GTL
