Leaving a registration number in a will

From a DVLA perspective I would imagine that you’d need to fill out the boxes on the retention form and set the eventual recipient as the nominee so that they could send the form off and assign the number to a vehicle.
From a practical perspective it would probably be easier and safer to assign the numbers to vehicles and leave the vehicles in the will.
A left of field choice - if you don’t need the numbers now why not gift them now and let the recipients assign them to their cars?

The right to display a registration number is treated the same way as other more tangible assets bequeathed in a will and Executors are obliged to obtain a market valuation of the registration on death and include that value in the deceased estate.
If the OP has no use for the numbers then gifting them now to the intended recipients as a potentially exempt transfer will in 7 years time effectively remove them from the value of OPs estate on death which will reduce the inheritance tax bill.
