my brother has a car that he isn't using at the moment and that he has just removed his insurance cover for, though it still has MOT and tax.

I have a comp policy for my own car that allows me to drive other cars, and i need to go on a round trip of about 50miles tomorrow (my car is not here for me to use).

Can i drive his car that he isn't using (it has MOT and tax) under my own comp policy, utilising the 3rd party allowance for other cars??

I think some policies say the other car must be insured by someone else. Otherwise, if you park it anywhere it won't be insured.

I believe that as long as the car is taxed and MOT'd all is OK - but it's probably worth mentioning to you insurance company for a long(ish) trip.

Just check your own policy document. If it says that you may do so, i.e. 'Drive a vehicle not owned by you or hired to you under a hire-purchase agreement', then it's O.K. so long as no exclusions or conditions apply to that clause. However, if you were to park it on the public highway, the owner could be guilty of having an uninsured vehicle on the public highway. So, if you will be able to park it off the public road at journey's end, you should be fine.
