I'm planning a day out for my good wife's birthday and I'm looking to get through the Cotswolds (which I know quite well) before ending up in The Chilterns, which I don't.

Can anyone recommend some nice driving routes and places to see or visit?

The Cotswolds encompasses a lot of territory. Where are you starting from?

Then I'll warn you now, the A4155 is bloody horrible!

Having grown up in the Chilterns, I can't think of many fun roads except the A40 when it climbs up the Stokenchurch cutting next to the M40. There are many 50 limits. The roads are bendy and full of villages with extended 30 limits out in to the country. Especially in Oxon.

The route I suggested already included the A40 up Aston Hill. From there to Henley only uses 3.5 miles of the A4155.
If avoiding even that is deemed necessary then go via Christmas Common followed by Stonor or Nettlebed.
