Recommendations for good roads in The Chilterns.

I'm planning a day out for my good wife's birthday and I'm looking to get through the Cotswolds (which I know quite well) before ending up in The Chilterns, which I don't.

Can anyone recommend some nice driving routes and places to see or visit?
